Characterization and Optimization of Hydrogel by Freezing-Thawing Method using Response Surface Methodology

Abstract
In this study, PVA-based physically cross-linked hydrogels were prepared by changing freezing time, the content of polymer and gelling agent using freezing (?40¡É) / thawing (25¡É, 3hr) method. The purpose of this study is to evaluate and optimize the formulation of hydrogel based on the design of experiment (DoE) using central composite design. Melting point and moisture content of hydrogels for 3hr or more freezing time were uniform. SEM results was showed cross-linked bridge and the incorporation of dexibuprofen. On the basis of results, springiness and drug content were not affected by independent variables. AS the content of PVA and PEG increased, the hardness and peak stress increased, but the moisture content decreased. Also released drug % was increased until 10% of PEG/PVA and decreased by more than 10%. By the results we can confirm that the mechanism of drug release from hydrogel is Kormeyer-Peppas model by non-fickian diffusion. Optimized hydrogel (PVA 8.97%, PEG 12.28%) showed satisfactory results for dependent variables.
